Exploration of defensive protein Amylase Trypsin Inhibitors from wheat: A novel approach for crop protection

نویسندگان

چکیده

Insect pests are highly dependent on starch-rich cereals and cause severe damage to the cereal grain nutrient yield. Amylase trypsin inhibitor protein degrades digestive enzyme alpha-amylase which plays a key role in carbohydrate metabolism as well growth development of insects. These proteins mainly found crops like wheat, maize, barley rich sources starch. Due defensive mechanism against pests, is prominent candidate for pest management crops. It could be used marker-assisted plant breeding genome mapping. Wheat amylase trypsin inhibitors drive intestinal inflammation via activation of toll-like receptor 4

Ingestion of wheat, barley, or rye triggers small intestinal inflammation in patients with celiac disease. Specifically, the storage proteins of these cereals (gluten) elicit an adaptive Th1-mediated immune response in individuals carrying HLA-DQ2 or HLA-DQ8 as major genetic predisposition. This well-defined role of adaptive immunity contrasts with an ill-defined component of innate immunity in...

Protease inhibitors in crop protection from insects

World wide crop losses without the use of pesticides and other non-chemical control strategies is estimated to be about 70% of crop production, amounting to U.S. $ 400 billion. Many insect pest families are known which cause serious damage to agriculture crops. Insect pest menace is one of the major factors that destabilize crop productivity in agricultural ecosystems. They are responsible for ...

Signal peptide homology between the sweet protein thaumatin II and unrelated cereal alpha-amylase/trypsin inhibitors.

A cDNA clone (pUP-23) corresponding to a member of a protein family that includes inhibitors of trypsin and of heterologous alpha-amylases has been selected from a library derived from developing barley endosperm and its sequence has been determined. A stretch of 95 nucleotides that included the signal peptide and the first 8 residues of the mature protein was found to be homologous to an exact...

Trypsin-specific inhibitors from the basidiomycete Clitocybe nebularis with regulatory and defensive functions.

We have isolated serine protease inhibitors from the basidiomycete Clitocybe nebularis, CnSPIs, using trypsin affinity chromatography. Full-length gene and cDNA sequences were determined for one of them, named cnispin, and the recombinant protein was expressed in Escherichia coli at high yield. The primary structure and biochemical properties of cnispin are very similar to those of the Lentinus...
